This is your choice. Most people opt for a slab, but if you want a wooden floor you would have to put down concrete footings.
Build up two courses of blocks, set your floor joists into the blocks, then pour concrete around them.
The wooden floor is then fixed down to the floor joists, and the Thermacell blockwork can then be continued up to the ceiling and filled in the normal way.
